Test tube strains are usually used for mother strains, and culture plate strains
(plate strains) are also used. Plate strains can be used to judge the quality of
strains by observing the colony morphology and mycelial growth rate. Manufacturers
that use test tube mother strains for a long time can regularly inoculate plates to
judge the vitality of mycelium.
The formulation and sterilization method of plates are the same as those of test tubes.
Usually, the culture medium and empty plates are sterilized separately. After the culture medium is
sterilized,It is dispensed into sterilized empty plates while hot in the clean bench. Each plate is
dispensed with 25~30 of culture medium and used after cooling. After the plate inoculation is
completed, the plate mouth is sealed with sealing film and placed in a biochemical incubator
for stacking and culture, with 5~6 plates placed in each stack. Pay attention to the temperature
and humidity of the incubator. After the plate culture is completed, a consistency evaluation is
carried out, and qualified strains are selected for use. Plate strains are not recommended for
short-term storage.
