Sterilization of Enoki Mushroom Cultivation Bottles (Bags)

After filling, the enoki mushroom cultivation bottles (bags) are placed in a reusable basket and placed on a sterilization cart. The key to successful factory cultivation is thorough sterilization; otherwise, all efforts are in vain. Sterilization containers include atmospheric pressure autoclaves, high-pressure autoclaves, and vacuum autoclaves. High-pressure autoclaves and vacuum autoclaves are generally used in factories.


(I) Sterilization

Sterilization requires sufficient time and thoroughness. The following sterilization guidelines are explained using a vacuum autoclave (9,600 bottles per pot) as an example for your reference.

1. Sterilization Procedure

Check the sterilization procedure → Verify all valves and switches → Push the cart into the sterilizer → Verify the quantity → Close the door → Verify the door seal → Start the sterilization procedure → Record → Verify temperature changes. 

2. Sterilization Specific Operation Requirements

① Open the cabinet door and ensure that both doors cannot be opened simultaneously.

② After loading bottles onto the cart, slowly push the sterilization cart into the sterilizer and arrange them neatly from the inside out.

③ Confirm that 24 carts (25 bottles per cart, 16 bottles per cart) are placed in the sterilizer and close the sterilization valve.

④ Confirm that the cabinet door is fully closed and sealed.

⑤ Confirm that the sterilization program settings are: sterilization pressure of 0.15 MPa, 105°C hold for 30 minutes, 110°C hold for 30 minutes, effective sterilization temperature of 122°C, hold for 80 minutes, and hold for 20 minutes.

⑥ Start the automatic sterilization program and confirm the temperature rise in the sterilizer.

⑦ The temperature rise in the sterilizer is 100°C, and the cabinet pressure is positive. Before maintaining pressure, exhaust all cold air from the autoclave. When the pressure rises from 0 MPa to 0.05 MPa, begin exhausting cold air to reduce the pressure to 0 MPa. Then, increase the pressure to 0.1 MPa and then vent the air until the pressure drops to 0 MPa. This venting process must be repeated at least twice.

⑧ Confirm that the sterilizer temperature is 105°C and maintain it for 30 minutes.

⑨ Automatically run the sterilization program, raising the temperature to 122°C, maintaining it for 80 minutes, and then allowing it to sit for 20 minutes.

⑩ Automatically run the sterilization program until completion, keeping relevant records.
