The stock is the most critical link in cultivation. The quality of the stock directly affects the yield. For stock production, excellent strains should be selected, including those selected by oneself or imported from other places.
1. Commonly used slant culture medium formula for stock
(1) Potato dextrose medium (PDA) 200g potatoes (peeled), 20g glucose, 18~20g agar, pH 6.5~7.0, 1000ml water.
(2) Potato comprehensive culture medium: 200g potatoes (peeled), 20g glucose, 18-20g agar, 3g potassium dihydrogen phosphate, 1.5g magnesium sulfate, pH 6.5-1.0, 1000ml water.
(3) Enoki mushroom rejuvenation medium: 200g potatoes (peeled), 20g glucose, 58% peptone, 18-20g agar, 3g potassium dihydrogen phosphate, 1.5g magnesium sulfate, pH 6.5-7.0, 1000ml water.
2. Preparation of mother culture medium: Prepare according to conventional methods.
3. Propagation of mother culture medium:Inoculate the mother culture medium with strong and vigorous mycelial growth and no infection into a sterilized blank agar slant medium in a sterilized aseptic operation, and then culture in an incubator at about 23°C. After 7-12 days, it becomes the mother culture medium used in production. At the desired temperature, the mother culture of Enoki mushrooms is prone to fruiting bodies, so the mother culture test tubes used for production need to be expanded into stock culture in a timely manner.
(III) Preparation of stock culture and cultivated culture (solid culture)
1. Common formulas for culture medium for stock culture and cultivated culture
(1) Sawdust culture medium: 78% sawdust, 20% wheat bran, 1% calcium carbonate, 1% sucrose.
(2) Cottonseed hull culture medium: 88% cottonseed hull, 10% bran, 1% calcium carbonate, 1% sucrose.
(3) Wheat grain culture medium: 99% wheat grain, 1% calcium carbonate.
2. Selection of culture bottle and bag: Standard 750ml culture bottle is commonly used for stock culture, and polypropylene bag with a length of 17cm, a width of 33cm, and a thickness of 0.004cm is generally used for cultivated culture.
